NEST Cell Culture Inserts with membrane PET (polyester) about pore size 0,4 μm are advanced cell culture systems designed to create reliable models of biological barriers and to study permeation and transmembrane transport vitroThese solutions are widely used in pharmacokinetic and toxicological studies, as well as in epithelial and endothelial models such as Caco-2 and MDCK. Inserts are available in 6-well, 12-well, and 24-well formats and are supplied with a matching multi-well cell culture plate, which ensures system stability, correct insert seating, and high experimental reproducibility. The PET membrane is characterized by very high optical transparency, enabling precise microscopic observations and fluorescence analyses. The "nested inserts" design guarantees uniform access to the medium in the upper and lower chambers and full compatibility with serological pipettes and automation systems.

0,4 μm PET membrane – tight barrier models and permeability control

Membrane PET with a pore size of 0,4 μm was designed for applications requiring high cell monolayer tightness. This configuration enables the creation of stable barrier models, where cell integrity control, substance transport studies, and permeability measurements, including TEER tests, are crucial. The uniform pore structure ensures repeatable diffusion conditions, minimizing uncontrolled flow between chambers. The high transparency of the PET membrane facilitates observations under brightfield and fluorescence microscopy, which is particularly important in the analysis of cell morphology and barrier markers. To obtain stable and comparable results, it is recommended to work with media and buffers prepared using sterile PES filters 0,22 μm.

Sterility, quality and laboratory standards

NEST inserts with membrane PET 0,4 μm are manufactured from virgin polystyrene of the highest class USP VI, which guarantees their biological safety and stability in long-term cell cultures. The insert surface is available in the following variants: TC-Treated, supporting the adhesion and growth of most adherent cell lines and facilitating the formation of a homogeneous barrier monolayer. Each insert is individually packaged in sterile Tyvek® blisters and sterilized using the E-Beam (SAL 10⁻⁶), ensuring the highest level of microbiological purity. The products are free of DNases, RNases, endotoxins and human DNA and meet the requirements IVDR 2017/746To maintain the sterility of the system and high repeatability of results, it is recommended to use sterile pipette tips with filter at every stage of work.
