LB Broth – a universal medium for culturing bacteria
LB Broth is a classic, liquid culture medium in the form of a dry powder, intended for general bacterial cultivation, in particular for the multiplication and short-term storage of strains Escherichia coli used in molecular biology, microbiology, and biotechnology. The medium is available in 250g bottles, allowing for the preparation of larger quantities of medium depending on the current laboratory needs and ensuring economical and flexible operation for both routine and research applications. For the inoculation and further cultivation of bacterial cultures, we recommend sterile petri dishes available from Googlab Scientific.
Composition and preparation of LB medium
The LB Broth formula is based on a proven and widely used formula, including 10 g of tryptone, 10 g of sodium chloride, and 5 g of yeast extract per liter of prepared medium, with a final pH of 7,0 ± 0,2 (25°C). To prepare the medium, weigh 25 g of powder, add distilled or deionized water to 1 liter, and then heat to boiling until the ingredients are completely dissolved. After pouring into flasks or test tubes, the medium is autoclaved at 121°C for 20 minutes, obtaining a clear and stable broth that promotes reproducible bacterial growth conditions. The following are also helpful in everyday laboratory work: sterile loops individually packed, facilitating the inoculation of cultures.
Quality control and storage conditions
Every batch of LB Broth is quality controlled using reference strains Escherichia coli, including ATCC 25922 and DH5α, incubated at 36 ±1°C for 12–18 hours. The acceptance criterion is the achievement of a characteristic, uniform turbidity in the broth, confirming the proper culture properties of the medium. The medium should be stored at 2–30°C, in the original, tightly closed packaging, protecting the powder from moisture. The shelf life is 36 months from the date of production, which guarantees stability and repeatability of culture parameters throughout the period of use.
