SB Broth – a rich medium for intensive bacterial cultivation

SB Broth is a dry, powdery culture medium intended for the rapid and efficient multiplication of bacteria, especially strains Escherichia coli used in molecular biology and genetic engineering. Thanks to the very high content of tryptone and yeast extract, the medium provides cells with significantly more nutrients than classic LB media, enabling the achievement of high cell density in a short time. The product is available in 250g bottles and is ideal for protocols requiring intensive bacterial growth. For safe inoculation of cultures, we recommend sterile loops individually packed available from Googlab Scientific.

Composition and preparation of SB Broth substrate

The SB Broth formula is based on a rich and precisely selected composition, including 32,0 g of tryptone, 20,0 g of yeast extract, and 5,0 g of sodium chloride per liter of prepared medium, with a final pH of 7,0 ± 0,2 (25°C). To prepare the medium, weigh 57,0 g of powder, dissolve it in distilled or deionized water, and then heat with stirring until completely dissolved and boiling. After pouring into appropriate containers, the medium is sterilized in an autoclave at 121°C for 20 minutes, obtaining a clear and stable medium that supports intensive bacterial growth.

High efficiency in plasmid and protein extraction

SB Broth was designed for applications requiring high bacterial biomass for downstream workflows, such as plasmid extraction and protein expression and isolation. In comparative tests with media based on raw materials from other manufacturers, Biologix medium provided higher average yields of plasmid DNA from E. coli DH5α (pUC19), while maintaining very good purity parameters, confirmed by A260/280 and A260/230 ratios. This makes SB Broth an optimal choice for laboratories conducting cloning, protein expression, and advanced molecular biology projects where reproducibility and high-quality results are crucial.
