TB Broth – a medium recommended for protein expression in Escherichia coli

TB Broth is a dry, powdery culture medium intended for high-yield protein expression in strains Escherichia coli used in molecular biology and genetic engineering. Thanks to its optimized, rich composition, the medium allows for significantly higher cell density and higher protein levels compared to standard culture broths. TB Broth is recommended for various expression induction methods, including those using IPTG, arabinose, or maltose, as well as for the resuscitation and storage of lyophilized bacterial strains. For safe inoculation of cultures, we recommend sterile loops individually packed available from Googlab Scientific.

TB Broth Composition and Medium Preparation

The TB Broth formula is based on a rich set of nutrients, including 12,0 g of tryptone, 24,0 g of yeast extract, 2,3 g of monopotassium phosphate (KH₂PO₄) and 12,5 g of disodium potassium phosphate (K₂HPO₄) per 1 liter of ready-made medium, at a final pH of 7,2 ± 0,2 (25°C). To prepare the medium, weigh 50,9 g of powder, dissolve it in distilled water while heating, then add 4 ml of glycerol, pour the medium into suitable containers and sterilize in an autoclave at 121°C for 15 minutes, obtaining a stable and highly buffered medium that supports intensive bacterial growth.

Performance, quality control and storage conditions

TB Broth has been validated in comparative tests with TB media prepared from raw materials from other manufacturers, under the same strain cultivation conditions. Escherichia coli DH5α with the pUC19 plasmid. Biologix medium provided higher average nucleic acid yields while maintaining very good purity parameters, confirmed by A260/280 and A260/230 ratios, confirming its suitability for demanding applications related to protein expression and plasmid work. The medium should be stored at 2–30°C in its original, tightly closed packaging, protecting the powder from moisture. It has a shelf life of 36 months from the date of production.
